Description
It is widely known that over-expression of translation initiation factors (eIF4E, eIF4G, eIF2, etc.) induces malignant transformation of NIH3T3 cells. The main question remaining to be answered is: What are the target genes affected by over-expression of these factors?. We have hypothesized that the immediate early effect of over-expression of these factors should affect the translation of a restricted group of transcripts, including the mRNAs responsible for the malignant transformation. To unravel these genes we over-express these factors in inducible vectors and are currently analyzing the expression profiles the polysome-bound transcripts ont these cells. This will allow us to identify the genes regulated at the translational level and implicated in the malignant transformation.
